چکیده
immunoglobulin, the clinical skin severity scores were lower in FcRg-/NC/Tnd mice by half than in conventional WT mice. On the other hand, there were no differences in both scratching behavior elicited by dermatitis and Th1/Th2 cytokine production between 2 groups of mice. In the skin lesion of FcRg null mice, mild epidermal hyperplasia and immune cell infiltration were observed. Particularly, mast cell numbers and their degranulation were significantly decreased in the skin of FcRg-deficient mice. Conclusions: FcRg was not critical to the onset of AD, because FcRg-deficient mice exhibited moderate dermatitis and scratching behavior comparable to WT. On the other hand, although scratching behavior induced the mechanical destruction of skin barriers and mast cell activation, the absence of FcRg markedly attenuated the skin severity, immune cell recruitment including lymphocytes, and mast cell degranulation. These results indicated that the FcRg-mediated immune response by specific IgG and IgE regulate the exacerbation of clinical symptoms in AD.

Successful treatment of anti-Caspr2 syndrome by interleukin 6 receptor blockade through tocilizumab.
IMPORTANCE A patient with a Caspr2 autoantibodies-associated syndrome had an unusual clinical triad and an excellent response to B-cell-anergizing therapy using the humanized monoclonal antibody tocilizumab directed against the interleukin 6 (IL-6) receptor. OBSERVATIONS A 55-year-old man had an atypical clinical triad of epilepsy, dysarthria, and paroxysmal kinesigenic dystonia, and a high t...
متن کاملTocilizumab for the Treatment of Rheumatoid Arthritis and Other Systemic Autoimmune Diseases: Current Perspectives and Future Directions
Interleukin (IL)-6 is a cytokine featuring redundancy and pleiotropic activity. While IL-6, when transiently produced, contributes to host defense against acute environmental stress, continuous dysregulated IL-6 production plays a significant pathological role in several systemic autoimmune diseases. In response to the expectation that IL-6 blockade would constitute a novel therapeutic strategy...
متن کاملAnti-Interleukin-6 Receptor Antibody Therapy-Induced Retinopathy in a Patient with Rheumatoid Arthritis
Tocilizumab, a humanized anti-human interleukin-6 (IL-6) receptor monoclonal antibody, is beneficial for treating autoimmune conditions such as rheumatoid arthritis (RA). The most common adverse event is upper respiratory tract infection; ocular side effects are rare. We describe a case of skin ulceration and bilateral retinopathy with multifocal cotton-wool spots and retinal hemorrhages in a p...
متن کاملTargeting Interleukin-6: All the Way to Treat Autoimmune and Inflammatory Diseases
Interleukin (IL)-6, a cytokine featuring redundancy and pleiotropic activity, contributes to host defense against acute environmental stress, while dysregulated persistent IL-6 production has been demonstrated to play a pathological role in various autoimmune and chronic inflammatory diseases. Targeting IL-6 is thus a rational approach to the treatment of these diseases. Indeed, clinical trials...
متن کاملMultifocal encephalopathy and autoimmune-mediated limbic encephalitis following tocilizumab therapy.
A 63-year-old man with rheumatoid arthritis developed multifocal encephalopathy and limbic encephalitis following therapy with tocilizumab, a humanized anti-interleukin-6 receptor antibody. Anti-glutamate receptor ε2 antibodies were later found to be positive in both the serum and cerebrospinal fluid. This case highlights the possibility of the development of encephalopathy after treatment with...
